摘要:
目的:探讨IgA肾病肾小球硬化模型的建立方法,为研究提供新的模型。方法:将24只SD实验大鼠随机分为假手术组、5/6肾切除组、SEB复合感染组和SEB复合感染合5/6肾切除组;12周末观察尿常规红细胞计数、24h尿蛋白、血清BUN及Scr情况;免疫荧光观察肾小球中IgA的表达;光镜和透射电镜分别观察大鼠肾组织结构及超微结构的情况;并运用免疫组化法检测TGF-β1在肾组织中的表达。结果:SEB复合感染联合5/6肾切除组尿常规RBC计数明显增多,并伴24h尿蛋白、血清BUN及Scr逐渐升高,肾小球系膜区可见大量IgA沉积,且强度较SEB复合感染组强,光镜和透射电镜可见大部分区域出现肾小球硬化伴系膜区电子致密物沉积,肾组织中TGF-β1表达明显上调。 结论:SEB复合感染联合5/6肾切除制作的IgA肾病肾小球硬化模型在一定程度上符合人类的临床病理特点,可提供一种新的模型。
Objective: To explore the method of establishment of IgA nephropathy with glomerulosclerosis, which provide a novel rat model. Methods: 24 male SD rats were randomly divided into four groups: Sham-operation group, 5/6 nephrectomy group, SEB infection group, SEB infection combined with 5/6 nephrectomy group. RBC count, urinary protein, serum BUN and Scr were detected at the end of 12th week. Immunofluorescence was employed to assay expression of IgA in glomerulus. Light microscope and transmission electron microscope were respectively used to analyse renal tissue structure and Ultrastructural structure. The expression of TGF-β1 in renal tissue was detected by immunohistochemisty. Results: RBC count in urinary sediment was the highest in the SEB infection combined with 5/6 nephrectomy group, with 24h urine protein, serum BUN and Scr increasing. Mass IgA deposits in glomerular mesangium were found in the SEB infection combined with 5/6 nephrectomy group, which is stronger than the co-infected group. Most ereas with glomerular sclerosis and were observed in the SEB infection combined with 5/6 nephrectomy group by Light microscope, and electron-dense matter was detected in mesangial region by transmission electron microscope, also up-regulation of TGF-β1 in renal tissue. Conclusions: IgA nephropathy with glomerulosclerosis was similar to clinicopathological feature using SEB infection combined with 5/6 nephroectomy, which can provided a novel model.
