实验性胰岛素抵抗2型糖尿病大鼠生物学特征研究

摘要:
目的:采用高脂饮食联合小剂量链脲佐菌素(streptozotocin, STZ)诱导胰岛素抵抗糖尿病大鼠模型,并对其生物学特征进行研究。方法:选择健康雄性SD大鼠20只,随机分为对照组(普通饲料)(10只)和模型组(高脂饲料)(10只),自由进食,4周后模型组大鼠腹腔注射STZ(35mg/Kg体质量),血糖≥16.67mmol/L者造模成功。造模成功大鼠继续进食高脂饲料。于成模后1周、4周及8周时观察各组大鼠的体质量、血糖、甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)和尿微量白蛋白(MAU)的变化,于成模后1周及8周时行IPGTT及ITT。结果:与对照组相比,模型组大鼠体质量在成模1周后明显升高(P<0.01),成模4周、8周时明显降低(p<0.01)。成模1周后,模型组大鼠血糖、TG、TC、LDL-c明显升高(p均<0.01),HDL-c降低(p<0.01),MAU升高(p<0.01); IPGTT和ITT实验结果显示模型组大鼠存在胰岛素抵抗。这些变化在整个实验过程中呈持续性。结论: 高脂饮食联合小剂量STZ诱导的糖尿病大鼠存在糖脂代谢紊乱及胰岛素抵抗。这种造模方法所诱导的伴胰岛素抵抗的大鼠糖尿病更近似于临床上的2型糖尿病的表型特征及发病过程,可用于2型糖尿病及相关慢性并发症的实验研究。
Objective: The Experimental Rat with Insulin Resistance and diabetes was induced by high fat diet and low dose Streptozotocin. Biochemical and metabolic characteristics of such rat model was studied in our experiments. Methods: Twenty male SD rats were randomly divided into two groups: experiment group fed with high-fat diet (n=10) and control group (n=10). After 4-weeks feed, the rats of high-fat diet-fed group were injected with low dose of streptozotocin of 35mg/Kg. Rats with plasma glucose over 16.67mmol/L were assigned to successfully type 2 diabetic model. The changes of body weight,glucose, triacylglyceride (TG), total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C) were measured at 1 week, 4 weeks and 8 weeks after the model was established. The intraperitoneal glucose tolerance test (IPGTT)and the intraperitoneal insulin tolerance test (ITT)in each group were compared at 1 week and 8 weeks after the model was established. Results: Compared to the normal rats, the diabetic rats which treated with high-fat diet and low-dose streptozotocin exhibited significant elevation of insulin resistance and high level of serum glucose, TC, TG, LDL-C and decreasing of HDL-C, which could be observed during the whole experiments. Conclusion: The diabetic rats induced by the high-fat diet and low-dose streptozotocin exhibited significant abnormality of glycolipid metabolism and elevation of insulin resistance which are closer to the pathophysiolgical changes of the type 2 diabetic patients and could be used in the experimental researches of type 2 diabetes and its chronic complications.
