摘要:
目的 观察大鼠双侧基底外侧杏仁核(BLA)脑区微量注射去甲肾上腺素(NE)对异丙酚遗忘效应的影响。 方法 将50只健康成年雄性Sprague-Dawley大鼠随机分为5组,每组各10只。S+S组:双侧BLA注射生理盐水+腹腔注射生理盐水;S+P组:双侧BLA注射生理盐水+腹腔注射25mg/kg异丙酚;NE0.3+P组:双侧BLA注射NE0.3μg +腹腔注射25mg/kg异丙酚;NE1.0+P组:双侧BLA注射NE1.0μg +腹腔注射25mg/kg异丙酚;NE3.0+P组:双侧BLA注射NE3.0μg+腹腔注射25mg/kg异丙酚。每组大鼠于连续被动回避实验(CMIA)训练前5min腹腔注射生理盐水或25mg/kg的异丙酚,于训练后即刻双侧BLA微量注射生理盐水或不同剂量的去甲肾上腺素,于训练后24h检测潜伏期及开场实验(OF)中的水平穿格次数及站立次数。 结果 与对照组S+S组相比,S+P组与NE0.3+P组训练后24h的潜伏期显著降低(P值均<0.05);NE1.0+P组与NE3.0+P组训练后24h的潜伏期与对照组相比差异无统计学意义(P值均>0.05)。各组间大鼠开场实验中水平穿格次数及站立次数差异均无统计学意义(P值均>0.05)。 结论 BLA去甲肾上腺素能系统参与了静脉麻醉药物异丙酚的遗忘效应。
Objective To investigate the effect of intra-basolateral amygdala infusions of norepinephrine on the amnesic effect of propofol in rats. Methods Fifty male adult Sprague-Dawley rats, weighing 250-280g, were randomly divided into 5 groups (n=10 each): S+S group rats received saline both intra-BLA and intraperitoneally; S+P group rats received saline intra-BLA and 25 mg/kg propofol intraperitoneally; NE0.3+P group rats received 0.3 μg norepineprine intra-BLA and 25 mg/kg propofol intraperitoneally; NE1.0+P group rats received 0.3 μg norepineprine intra-BLA and 25 mg/kg propofol intraperitoneally; NE3.0+P group rats received 3.0 μg norepineprine intra-BLA and 25 mg/kg propofol intraperitoneally. Rats were intraperitoneally injected with either saline or propofol (25mg/kg) 5 minutes before the training of continuous multiple-trail inhibiton avoidance (CMIA), and subsequently received bilateral infusions of saline or norepinephrine (0.3, 1.0, or 3.0 μg) into the BLA immediately after training. Twenty-four hours later, the CMIA retention latency and number of crossings and number of rearings of the open field test were tested. Results The retention latencies of S+P group and NE0.3+P group were significantly lower than S+S group (P<0.05). The retention latencies of NE1.0+P group and NE3.0+P group were restored to the baseline level (P>0.05). There were no significant differences in the number of crossings and number of rearings among the groups (P>0.05). Conclusion The noradrenergic system in the BLA is involved in the mechanisms of propofol amnesic effect.
