时辰对罗哌卡因、氯普鲁卡因和利多卡因惊厥ED50及LD50的影响

摘要:
目的 探讨时辰对罗哌卡因、氯普鲁卡因、利多卡因惊厥ED50及LD50的影响。方法 270只小鼠随机分为两组即ED50组和LD50组，每组135只；每组再分为3个亚组即罗哌卡因组、氯普鲁卡因组、利多卡因组，每组45只；按不同时辰每组再随机分为3个次亚组，每次亚组15只,分别于8∶00、14∶00和20∶00腹腔注射罗哌卡因、氯普鲁卡因、利多卡因，以小鼠惊厥和死亡为观察指标，用序贯法测定各时间点的ED50 及LD50。结果　利多卡因在14∶00的惊厥ED50低于8∶00和20∶00（P ＜0.05、P ＜0.01），其余各时辰ED50的差异无显著性(P ＞0.05)；利多卡因在14：00的LD50低于8：00（P ＜0.05）。罗哌卡因8∶00的惊厥ED50和LD50均低于14∶00和20∶00（P ＜0.05、P ＜0.01) ，其余各时辰的ED50及LD50的差异无显著性 (P ＞0.05)。氯普鲁卡因在8:00的ED50低于14：00（P ＜0.05），氯普鲁卡因各时辰LD50差异无显著性 ( P ＞0.05)。 结论　罗哌卡因、氯普鲁卡因和利多卡因惊厥ED50具有时辰依赖性。利多卡因、罗哌卡因的LD50具有时辰依赖性，氯普鲁卡因LD50未发现时辰依赖性。
Objective To observe the effects of chrono on the median effective dose(ED50 ) and the median lethal dose (LD50) of convulsion induced by anaesthetics (Ropivacaine,Chloroprocaine,Lidocaine ). Methods 270 mice were randomly assigned into 2 groups for LD50 and ED50 ,135 mice per group, which were then assigned into 3 Subgroups, 45 mice per subgroup. Each subgroup was assigned to 3 tertiary- groups,15 mice each .Ropivacaine,Chloroprocaine and Lidocaine were intraperitoneally injected at 8∶00 ,14∶00 and 20∶00 ,respectively. The convulsion was investigated and both ED50 and LD50 at different times were determined by up-down sequential allocation in mouse. Results The convulsive ED50 of Lidocaine at 14∶00 was lower than those at 8∶00 and 20∶00 (P ＜0.05、P ＜0.01) ,The LD50 of Lidocaine at 14∶00 was lower than that at 8∶00 ( P ＜0.05) .The convulsive ED50 and LD50 of Ropivacaine at 8∶00 were lower than those at 14∶00 and 20∶00（P ＜0.05、P ＜0.01）. The convulsive ED50 of Chloroprocaine at 8∶00 was lower than that at 14∶00 ( P ＜0.05).Few effects of chrono on the LD50s of Chloroprocaine had been investigated. Conclusion 　The effects of convulsive ED50s of Ropivacaine, Chloroprocaine and Lidocaine were chrono-dependent . The effects of LD50s of Ropivacaine and Lidocaine also were chrono-dependent . The effects of LD50s of Chloroprocaine were not investigated.
