营卫调节方对兔血管外膜损伤致内膜增生性病变的干预作用

摘要:
目的:探讨营卫调节方在兔颈动脉外膜损伤致动脉粥样硬化中的作用。方法:新西兰大白兔高脂喂养两周后,剔除血脂不高者,采用胶原酶消化+机械分离的方法建立兔单侧颈动脉外膜损伤模型,对侧行假手术。术后继续高脂饮食喂养,随机分为正常对照组、外膜损伤组以及营卫调和方+外膜损伤组,分别于术后1、2、4、8周处死,取目标颈动脉行HE染色观察动脉粥样硬化病变情况、免疫组化法检测血管局部病变成分,ELISA法检测组织及外周血中炎症因子水平。结果:高脂饮食下给予营卫调节方的新西兰兔颈动脉在外膜损伤后1周未见明显内皮排列紊乱及增生,外膜损伤后2周发生轻度增生性病变,外膜损伤后4-8周,增生性病变逐渐消退;SM-α-actin免疫组化染色提示,营卫调节方可以减轻外膜损伤后平滑肌细胞的迁移;营卫调节方还可以使血管局部及外周血中IL-6、TNF-α水平明显降低(P<0.05);结论:颈动脉外膜损伤结合高脂饲料饮食能建立动脉粥样硬化动物模型,营卫合剂治疗后可降低斑块面积以及体内炎症因子水平,抑制动脉粥样硬化的进展。
Abstract Objective: To explore the role of Ying and Wei accommodation herbal decoction in rabbit vascular adventitial injury caused intimal hyperplasia. Methods: New Zealand white rabbits were fed with high cholesterol diet about two weeks, then rejected those who are not hyperlipidemia, models of New Zealand rabbits with vascular adventitial injury were established by collogenase digestion and mechanical dissection. Continue feeding high cholesterol after operation, then divided into three camps randomly: normal control group, Ying and Wei accommodation herbal decoction treatment after adventitial injury group and adventitial injury group, put to death after1, 2, 4 and 8 weeks since operation, HE staining were used to detect the morphological changes in vessels after adventitial injury. Immunohistochemisty is used to evaluate pathological change. The change of inflammatory cytokines in organism and peripheral blood were detected by ELISA. Results: Under the high cholesterol diet, the rabbit giving Ying and Wei accommodation herbal decoction carotid artery adventitial have no obvious endothelial disorder and hyperplasia after 1 week since operation. Then, 2 weeks after adventitial injury, the rabbit carotid artery adventitial had a mild intimal hyperplasia. Until 4-8 weeks after adventitial injury, the intimal hyperplasia had gradually faded. The Ying and Wei accommodation herbal decoction can lighten the smooth muscle cell immigration after adventitial injury by SM-α-actin staining. The level of IL-6 , TNF-αin organism were obviously decreased (P<0.05), and which in peripheral blood have the same trend. Conclusion: Vascular adventitial injury with high cholesterol diet can establish models of animal artherosclerosis. Ying and Wei accommodation herbal decoction can reduce the plaque area, as well as levels of inflammatory factors in vivo, restraining the progress of atherosclerosis.
